LIFT 5 will travel along Route 135 from downtown Hopkinton through Ashland and into Southern Framingham, with several scheduled stops, including Collela's Supermarket in Hopkinton and Ashland High School. In downtown Framingham riders can take the MBTA Commuter Train into Boston, transfer to LIFTs 2, 3, 6, and 7, or continue on LIFT 5 to Framingham State College.

LIFT 6 will travel Route 16 from downtown Milford to downtown Holliston, and continue north on Route 126 into Ashland and downtown Framingham. Scheduled stops include Milford and Holliston Town Halls, the Park and Ride, K-Mart, and the Radisson Hotel in Milford, and Shaw's Supermarket and Market Basket Plazas in Ashland. In downtown Framingham, riders can take the MBTA Commuter Train into Boston or Worcester, transfer to LIFTs 2, 3, 5 and 7, or continue on LIFT 6 to the Natick Mall, Sherwood Plaza, Logan Express, and Shoppers World.

In addition to scheduled stops, LIFT buses will stop anywhere along their routes to pick up passengers, as long as it’s safe to do so. Residents can hail LIFT buses by waving.
